Only if MacPorts thinks that it's installed to /opt/local. If you build MacPorts from source with --prefix, as Ryan suggested, it should work regardless of whether you make a symlink later.
This could be useful if you don't want to put /Volumes/foo/bar/baz/opt/local/bin in $PATH, for instance.
