Comment:

 Does your pkg-config work? Try:

 {{{
 /opt/local/bin/pkg-config --libs glib-2.0
 }}}

 It should say something like:

 {{{
 -L/opt/local/lib -lglib-2.0 -lintl -liconv
 }}}

 Do you have anything in /usr/local or /sw? These can interfere with
 MacPorts and their presence while using MacPorts is not recommended.
