Comment(by blb at macports.org):

 When checking for the gtk2 version installed, it does show the 2.14.3
 version as being active right?  As in:
 {{{
 $ port installed gtk2
 The following ports are currently installed:
   gtk2 @2.12.9_0+x11
   gtk2 @2.14.3_3+x11 (active)
 }}}
 If an older one is active, that would be the problem.

 Also, note there is no port py25-glib but py25-gobject.
