 I had exactly the same problem on my Mac OS X 10.5.8.  I tried to re-
 install MacPorts after removing /opt, but still failed to build.  Then I
 found that X11 was not installed or removed for some reason, which caused
 the problem.  After I've installed XQuartz 2.5.0, I was able to build and
 install py26-matplotlib correctly.
