 Works for me, OSX 10.11.6 El Capitan, vis:-
 {{{
 sudo port install x264 +universal configure.args.i386="--host=i686-apple-
 darwin"
 }}}
 I understand x264 sees El Capitan and guesses target is x86_64-apple-
 darwin, despite +universal, so tries to asm instructions that can't (at
 best).  By explicitly appending --host=i686-apple-darwin to the i386
 slice, we keep the optimised x86_64, and get the best i386 too.\\
 The above command line is a workaround until the port is upgraded.  Many
 thanks for the ticket :-)
