Joshua Root just answered this same question for some one else. He  
wrote:
MacPorts is installed into /opt/local, so to use the port command, you
need to have /opt/local/bin in your PATH. See
<http://guide.macports.org/#installing.shell>.

The installer for MacPorts 1.7 will do this for you more reliably.
