10.6 XQuartz, yes.  It shouldn't interfere with anything in MacPorts,
although launchd may be whining a bit if the launchagent file is still there
(/Library/LaunchAgents/org.macosforge.xquartz.startx.plist,
/Library/LaunchDaermons/org.macosforge.xquartz.privileged_startx.plist).
